   Sure, in the sense of:

http://dictionary.cambridge.org/dictionary/british/knock-off-sth
to stop working, usually at the end of the day
I don't knock off until six.
What time do you knock off work?
(Definition of knock off (sth) from the Cambridge Advanced Learner's Dictionary)

"let's knock off early"

Vamos acabar (o trabalho) mais cedo.

He meant somethig like "do jeito que tiver tá bom, assim vamos embora mais cedo."

He meant that the "asses" were made to perfection, not a sloppy job, if it was the maker would knock off work early.

It´s worth noticing that is a sitcom with their ironic/funny way of speaking.
