In that case it´s not a noun, it´s a verb and you would use the gerund form.

The question is "when is your sister graduating?" - quando a sua irmã vai se formar?

If was a noun, the question would be "when it´s your sister´s graduation?"  or "when is the graduation of your sister?". The apostrophe would mean "of".

The "is" (verb to be) along with the absence of apostrophe (or the preposition "of") suggests we need a verb to complete the meaning. So "graduating" would be the word there.
